science and were first expounded as such by Russell and Burch in 1959.
They may be presented as follows:
replacement: the use of non-animal methods such as cell cultures,
human volunteers and computer modelling instead of animals to achieve a

reduction: the use of methods that enable researchers to obtain
comparable amounts of information from fewer animals, or more.
refinement: the use of methods that minimise potential pain,
suffering or distress, and that enhance animal welfare for those animals
that cannot be replaced.
Hardware and software

The PV loops analyzer of iox2 software performs analysis on loops during steady-state and changing conditions (e.g. vena cava occlusion). Parameters from steady-state loops are calculated in real-time on a beat-by-beat basis. The analyzer processes occlusion loops (selected by user) to generate several parameters and graphs: end-systolic pressure-volume relationship (ESPVR); end-diastolic pressure-volume relationship (EDPVR); preload recruitable stroke work (PRSW); dPdtmax vs end-diastolic volume relationship; maximal elastance... The pressure and volume signals are obtained using a conductance catheter and signal conditioner. Millar hardware is available from emka TECHNOLOGIES. The robot lights cigarettes then ejects them when they are finished. Up to 24 cigarettes can be 'smoked' in a user-defined order - any combination of sequential or parallel smoking is possible.

The O.P.T. (oscillometric pressure telemetry) module is available as an option with the emkaPACK telemetry system. Our method of measuring blood pressure offers several valuable advantages, including:

The system is undergoing testing by a CRO. Preliminary results were presented recently at the 2009 meeting of the Society of Toxicology in Baltimore; they show that, after 1000 hours of recording from 40 dogs, reliable data were produced from 60% to 90% of inflation/deflation cycles.

The ecgTUNNEL range from emka TECHNOLOGIES can be used to measure 6-lead ecg in a non-invasive manner from conscious mice or rats. Mouse models have already been launched and are in use by customers. To satisfy additional requests, we developed the rat models.

The primary use is in inhalation experiments in rats and mice, using a whole-body chamber or a nose-only mask. However inExpose is also used in experiments with large animals (face mask) and tissue cultures.
It can be used for exposure to aerosolized liquids, suspensions and powders, toxins, vapors, gases… Notably, several accessories have been developed to meet the varying needs of researchers studying the effects of cigarette smoke.

Animal cages are adapted to receive food and drink holders. Each holder is fitted with a force transducer that is connected to iox acquisition software - up to 32 cages can be monitored simultaneously.
A specific iox analyzer quantifies intake, providing answers to questions like "How many times does the subject eat/drink during the day and during the night?" or "How much food/ drink is consumed each time?"
The analyzer also provides parameters related to visits without intake (for example subject poking around), so that other behavioural patterns may be detected.

automatic beat selection in iox

"Automatic beat selection" is a new feature in iox, data acquisition and real-time analysis software from emka TECHNOLOGIES.
iox selects beats using a protocol created by the user. A protocol consists of a set of instructions, each of which selects a single beat in relation to user-defined experimental marks (periods and comments). For example, "select a beat at 2 minutes after the start of the period named Low Drug Concentration".
Our software engineers primarily developed automatic beat selection for researchers performing electrophysiological studies: the ability to select action potentials using a protocol is a valuable time-saving device.
Selected beats are transferred to the Beat report panel, where changes are visible at a glance. In the screenshot below, we can see that the drug product has a clear effect on the ventricular action potential.

The microvessel myograph from emka TECHNOLOGIES is designed for the study of vessels with a diameter of 100µm to 2mm and a maximum length of 3.5mm. The transducer measures contractile forces of up to 10g.
Two models are available, with 1 bath (20ml) or 2 baths (2 x 3ml, 2 x 5ml, or 2 x 10ml).
emka TECHNOLOGIES can supply all that is necessary to start running experiments, including an amplification system, heater/pump for heating fluid, acquisition software and carbogen bottles. The carbogen provides the necessary oxygenation and also "pushes" the physiological solution into the baths (i.e. the system does not rely on a pump or the force of gravity for this purpose). A vacuum source is also required - if your laboratory does not have one, our technicians can install a vacuum pump with automatic switch.

Testing showed that data acquired from rabbits were of comparable high quality: 4 electrodes and 2 belts were connected to the emkaPACK for the acquisition of 6-lead ecg, lung volume and activity parameters. Signals could be processed without difficulty in iox and ecgAUTO.

Setups for measuring non-invasive blood pressure (nibp) are based on the principle of blood flow occlusion using a cuff placed around the subject's tail.emka TECHNOLOGIES offers CODA systems (Kent Scientific Corp.) for measuring the following nibp parameters from rats or mice:

| The ecgTUNNEL, consisting of a restrainer and electrodes, is a method for obtaining ecg recordings from conscious mice and other small rodents.
A cylinder-shaped tube restrains the subject firmly and gently, without causing trauma. Four electrode pads on the floor of the ecgTUNNEL allow you to obtain 1- to 6-lead ecg data.

The mainframe houses a card for 6 standard ecg leads and, optionally, a card for 6 precordial ecg leads, allowing for the acquisition of 6- or 12- lead ecg.
The mainframe connects directly to the acquisition card on the PC, eliminating the need for an interface box. Given that 12 channels are reserved for ecg acquisition; channels 13-16 can be used for the acquisition of other signals as desired (via bnc connectors on the mainframe).
To satisfy glp requirements, there are no connectors or user accessible settings on the cards (only a switch to select calibration or measurement modes).

emka TECHNOLOGIES provides complete hardware and software solutions for plethysmography studies with anesthetized subjects, for real-time direct measurement of airway mechanics - resistance and compliance - as well as standard respiratory parameters.
Subjects may be ventilated or breathing spontaneously. The standard hardware includes a whole-body plethysmograph fitted with a pneumotachograph, two differential pressure transducers and bridge amplifiers.
At the software end, real-time analysis is done in emka TECHNOLOGIES data acquisition software, iox2, with the rc (respiratory compliance) analyzer.
Add-on options allow for delivery of aerosol and/or anesthetic gas to the plethysmograph. The nebulization system for aerosol delivery is a complete system (aerosol generator, nebulization controller device, bias-flow pump etc), which integrates fully into the plethysmography setup.

What you should know before you design, build and equip your facility to conduct experiments with large populations using non-invasive telemetry.
When planning studies involving a high number of telemetered subjects, and hence a large experimental area, an obvious design consideration is hardware infrastructure. For example, effective management of several telemetry devices requires a single remote center from which the entire system can be monitored, controlled and operated. The architecture must also allow for distant computers to perform tasks such as: recording start and stop; real-time data control and visualization; hardware monitoring; data post processing; review and editing; database management; backup and archiving services; secured links to in-house LIMS. And all of this bearing in mind the large volume of data involved, especially now that video is frequently added to physiological data.
These requirements, compounded by obligations to comply with GLP and internal IT rules, make the choice of an ideal system all the more difficult. Large telemetry projects require in-depth case-by-case engineering design based on novel and powerful data and network management technologies.

double-chamber plethysmograph now available for mice!

Did you know? emka TECHNOLOGIES has developed a double-chamber plethysmograph to investigate pulmonary function in mice, before and after exposure to a drug or other challenge. The new size was adapted from the existing double-chamber plethysmograph for rats and guinea-pigs, and offers the same possibilities.
A neck-seal divides the plethysmograph into two independent chambers, for the subject's head and the rest of its body. The direct measurement of respiration in the head chamber allows absolute values to be obtained.
The double-chamber plethysmograph for mice may also be used with the body chamber only ("head-out" configuration).
